Cross-modality Neurotoxicity Prediction Using Human CNS-3D Organoids and Machine Learning

Accurate neurotoxicity prediction remains a key challenge in drug development. To validate CNS-3D Brain Organoids as a preclinical neurotoxicity model, 150 small molecules and a set of ASOs were assessed using functional neurophysiological endpoints and analyzed with an XGBoost machine-learning model. Across this dataset, CNS-3D showed significantly improved sensitivity and specificity relative to animal models, supporting their use for clinically predictive neurotoxicity assessment.
